Gene therapy for hemophilia B mice with scAAV8-LP1-hFIX

Hemophilia B is a hemorrhagic disease caused by the deficiency of clotting factor IX (FIX). Gene therapy might be the ultimate strategy for the disease. However, two main problems that should be solved in gene therapy for hemophilia B are immunity and safety. Self-complementary adeno-associated virus serotype 8 (scAAV8), a non-human primate AAV featuring low immunogenicity and high transfection efficiency in liver cells, might be a potential vector for hemophilia B gene therapy. A strong liver-specific promoter-1 (LP1) was inserted and mutant human FIX Arg338Ala was introduced into plasmid scAAV8-LP1 to develop an optimized AAV8 vector that expresses human clotting factor FIX (hFIX). The efficiency of scAAV8-LP1-hFIX administered through normal systemic injection or hydrodynamic injection was compared. A high expression was achieved using hydrodynamic injection, and the peak hFIX expression levels in the 5×1011 and 1×1011 virus genome (vg) cohorts were 31.94% and 25.02% of normal level, respectively, at 60 days post-injection. From the perspective of long-term (200 days) expression, both injection methods presented promising results with the concentration value maintained above 4% of normal plasma. The results were further verified by enzyme-linked immunosorbent assay and activated partial thromboplastin time. Our study provides a potential gene therapy method for hemophilia B.

Prediction of Driver Gene Matching in Lung Cancer NOG/PDX Models Based on Artificial Intelligence Article

Patient-derived tumor xenografts (PDXs) are a powerful tool for drug discovery and screening in cancer. However, current studies have led to little understanding of genotype mismatches in PDXs, leading to massive economic losses. Here, we established PDX models from 53 lung cancer patients with a genotype matching rate of 79.2% (42/53). Furthermore, 17 clinicopathological features were examined and input in stepwise logistic regression (LR) models based on the lowest Akaike information criterion (AIC), least absolute shrinkage and selection operator (LASSO)-LR, support vector machine (SVM) recursive feature elimination (SVM-RFE), extreme gradient boosting (XGBoost), gradient boosting and categorical features (CatBoost), and the synthetic minority oversampling technique (SMOTE). Finally, the performance of all models was evaluated by the accuracy, area under the receiver operating characteristic curve (AUC), and F1 score in 100 testing groups. Two multivariable LR models revealed that age, number of driver gene mutations, epidermal growth factor receptor (EGFR) gene mutations, type of prior chemotherapy, prior tyrosine kinase inhibitor (TKI) therapy, and the source of the sample were powerful predictors. Moreover, CatBoost (mean accuracy = 0.960; mean AUC = 0.939; mean F1 score = 0.908) and the eight-feature SVM (mean accuracy = 0.950; mean AUC = 0.934; mean F1 score = 0.903) showed the best performance among the algorithms. Meanwhile, application of the SMOTE improved the predictive capability of most models, except CatBoost. Based on the SMOTE, the ensemble classifier of single models achieved the highest accuracy (mean = 0.975), AUC (mean = 0.949), and F1 score (mean = 0.938). In conclusion, we established an optimal predictive model to screen lung cancer patients for NOD/Shi-scid, interleukin-2 receptor (IL-2R) γnull (NOG)/PDX models and offer a general approach for building predictive models.

Valveless pumping using a two-stage impedance pump

Impedance pump is defined as a type of valveless pumping mechanism, where an elastic tube is joined with a tube of different impedance, a periodic asymmetrical compression on part of the elastic tube will produce a unidirectional flow. Numerical and experimental studies had been conducted in the past, but little experimental work was found to exploit the principle of multi-stage impedance pumping for fluid transportation. This paper concentrates on the fluid behavior for an open-loop two-stage system experimentally. The aim of this work is to have an insight on the deliverable head and pumping rate of an open-loop two-stage impedance pumping system. This paper generally investigates the effect of different parameters towards the generation of unidirectional flows and composite flow of two opposite directions. Results obtained are of significant values.
